Prediction of Aggregation Sites in Human Eye Lens γ-Crystallins: Implications in Cataract
نویسندگان
چکیده
Cataract, a leading cause of blindness, occurs due to aggregation of crystallin proteins in eye lens. In this paper, the aggregation site in three major γ-crystallins of human eye lens has been identified unambiguously by using their primary sequences with the help of PASTA, Tango and AGGRESCAN. We have found that some hydrophobic residues in C-terminal domain of γ-crystallins play putative role in initiation of aggregation of these proteins. Suppression of aggregation of γD-crystallin by molecular chaperone in lens like αB-crystallin is also documented mechanistically through docking studies.

Cataract-Causing Defect of a Mutant γ-Crystallin Proceeds through an Aggregation Pathway Which Bypasses Recognition by the α-Crystallin Chaperone
BACKGROUND The transparency of the eye lens depends upon maintenance of the native state of the γ- and β-crystallins, which is aided by the abundant chaperones αA- and αB-crystallin. Mature onset cataract, the leading cause of blindness worldwide, involves the polymerization of covalently damaged or partially unfolded crystallins into light-scattering aggregates. A number of single amino acid s...

Considerable mechanistic insight has been gained into amyloid aggregation; however, a large class of non-amyloid protein aggregates are considered “amorphous,” and in most cases little is known about their mechanisms. Amorphous aggregation of γ-crystallins in the eye lens causes a widespread disease of aging, cataract. We combined simulations and experiments to study the mechanism of aggregatio...

متن کاملAssociation of partially folded lens βB2-crystallins with the α-crystallin molecular chaperone
Age-related cataract is a result of crystallins, the predominant lens proteins, forming light-scattering aggregates. In the low protein turnover environment of the eye lens, the crystallins are susceptible to modifications that can reduce stability, increasing the probability of unfolding and aggregation events occurring. It is hypothesized that the α-crystallin molecular chaperone system recog...
متن کاملThe Thermal Structural Transition of Alpha-Crystallin Modulates Subunit Interactions and Increases Protein Solubility
BACKGROUND Alpha crystallin is an oligomer composed of two types of subunits, alpha-A and alpha-B crystallin, and is the major constituent of human lens. The temperature induced condensation of alpha-crystallin, the main cause for eye lens opacification (cataract), is a two step-process, a nucleation followed by an aggregation phase, and a protective effect towards the aggregation is exhibited ...
